The trio's zombie-blasting video game-inspired action film, Resident Evil: Extinction, beat off all competition over the weekend with an estimated $24 million (GBP12 million) take.
It marks a record for the Resident Evil franchise - Resident Evil: Apocalypse opened with $23 million (GBP11.5 million) in 2004.
Pregnant Jovovich has starred in all three films.
Jessica Alba comedy Good Luck Chuck debuted at number two with $14 million (GBP7 million) and last weekend's (16Sep07) number one, The Brave One, falls to three.
